I've tried to do clean delinations of color by enlarging the painting to
500+ and go pixel by pixel - but it still doesn't look as "clean" as
I'd
like it to -- are there any other suggestions on how to get a really
straignt (or curved) clean line in my paintings??

Since with pixels you are working on a square grid, there's no way to obtain
clean curved lines: eventually jaggedness should appear, which would be more
noticeable when zooming in. Anti-aliasing kind of fixes this, since it
places some semi-opaque pixels to the side of the pixel you are painting,
which optically smooths the line, but if done incorrectly can sometimes turn
it a little bit blurry.
